OPTICAL SYSTEM:

  • The optical system is comprised of light excitation source (lasers) of a specific wavelength, and optical filters and mirrors which direct specific wavelengths of light to their appropriate detector.

  • Flow cytometers have three or more lasers of different wavelengths. This provides for multiple fluorescent measurements simultaneously.

  • Several optical filters are arranged in front of each photomultiplier tube (PMT) to direct the desired wavelength of light to the detector.

  • Photodiodes are commonly used for strong signals, such as forward light scatter.

  • Both detectors convert light signal into an electrical signal that can be further processed and analyzed by computers
